Can I start my car once the spark plugs are installed without putting the manifold back on to make sure the new spark plugs are wired correctly?

Not a good idea. You could stuff stuff into your car if it runs. The manifold has a lot of sensors in it for air flow, vacuum to adjust fuel flow, etc to make the car run.

My 9/16 spark plug seems to fit can I use it i instead of 14 mm?

What do you all recommend for sealing the manifold back? Last time the seal disintegrated and inused permutex. I will probably get the oem seal.

Learn metric conversion...............9/16" and 14 mm. are the same.

I would warn you that there are many places where SAE sockets will round the corners off of metric bolts after they seem to fit.............why you do conversion, to see how close they come to being the same, off more than .005" and you are asking to ruin parts.
